Glimepiride

Type 2 DM.

Initially 1 mg once daily, may be increased after each 1-2 wk treatment. Max dose: 6 mg/day. Individualized dosage when used in combination therapy w/ metformin or insulin.

Should be taken with food: Take immediately before the 1st main meal of the day. Do not skip meals. Swallow whole, do not chew/crush.

Hypersensitivity to glimepiride, sulfonylureas or sulphonamides. Patients w/ type 1 DM; diabetic ketoacidosis; diabetic coma. Severe kidney & liver disease.

Patients recovering from injury, operation, infections w/ fever, or other forms of stress. Risk of haemolytic anaemia in G6PD-deficient patients. Increased risk of hypoglycaemia in case of undernourishment, irregular meal time, missed or delayed meal or period of fasting; changes in diet; excessive dosage intake; hormone-induced disorders (disorders of thyroid gland, pituitary gland or adrenal cortex); alcohol intake; increased body exercise & inadequate carbohydrate intake. Regularly monitor blood or urine sugar, blood cell levels & liver function. Contains lactose. May affect ability to drive & use machinery. Severe liver disease or kidney disorder. Pregnancy & lactation. Childn <18 yr.

Hypersensitivity of the skin (eg, itching, rash, hives & increased sensitivity to sun), allergic reactions (eg, inflammation of blood vessels often w/ skin rash) which may progress w/ breathing difficulties, fall in BP & shock; abnormal liver function (eg, jaundice, cholestasis, hepatitis or liver failure); severe hypoglycaemia (loss of consciousness, seizures or coma).

Increased blood sugar lowering effect w/ other antidiabetics (eg, insulin, metformin); NSAIDs (eg, phenylbutazone, azopropazone, oxyphenbutazone, aspirin-like medicines); drugs for UTI (eg, long acting sulfonamides); antibacterial & antifungal agents (eg, tetracyclines, chloramphenicol, fluconazole, miconazole, quinolones, clarithromycin); anticoagulants (coumarin derivatives eg, warfarin); anabolics; male sex hormone replacement therapy; antidepressants (eg, fluoxetine, MAOIs); antihypercholesterolemia (eg, fibrates); antihypertensives (eg, ACE inhibitors); antiarrhythmics (eg, disopyramide); antigout agents (eg, allopurinol, probenecid, sulfinpyrazone); antineoplastics (eg, cyclophosphamide, ifosfamide, trofosfamide); wt-reducing agents (eg, fenfluramine); agents used to increase circulation (eg, IV pentoxifylline); antihistamine (eg, tritoqualine); sympatholytics. Decreased blood sugar lowering effect w/ female sex hormones (eg, oestrogen, progestogens); antihypertensives (eg, thiazide diuretics); TSH (eg, levothyroxine); steroids (eg, glucocorticoids); antipsychotics (eg, chlorpromazine & other phenothiazine derivatives); adrenaline & sympathomimetics; antihypercholesterolemia (eg, nicotinic acid); laxatives; antiepileptics (eg, phenytoin); sedative-hypnotics (eg, barbiturates); anti-glaucoma (eg, acetazolamide); diazoxide; anti-TB (eg, rifampicin); glucagon. Altered blood sugar lowering effect w/ antiulcerants (eg, H₂ antagonists); β-blockers, clonidine, guanethidine, reserpine; alcohol. Altered effects of anticoagulants (coumarin derivatives eg, warfarin). Altered absorption w/ colesevelam.

Antidiabetic Agents

A10BB12 - glimepiride ; Belongs to the class of sulfonylureas. Used in the treatment of diabetes.
