Donepezil HCl
Indications/Uses
Symptomatic treatment of mild, moderate & severe Alzheimer's disease, vascular dementia (associated w/ CV disease) & dementia w/ Lewy bodies (DLB).
Dosage/Direction for Use
Adult & elderly 5 mg once daily. May be increased to 10 mg once daily after at least 1 mth. Max: 10 mg/day. Patient w/ DLB Dose may be decreased to 5 mg depending on the symptoms of the patient.
Administration
May be taken with or without food: Place tab on the tongue & allow to disintegrate then, swallow w/ or w/o water.
Contraindications
Hypersensitivity to piperidine derivatives. Pregnancy & lactation.
Special Precautions
Succinylcholine-type muscle relaxation during anesth. Sick sinus syndrome or other supraventricular cardiac conditions eg, SA or AV block. Patients at risk for developing ulcers, seizures. Increased incidence of worsening extrapyramidal disorder in patients w/ dementia w/ Lewy bodies. History of asthma or obstructive pulmonary disease. May impair ability to drive or operate machinery. Childn.
Adverse Reactions
Diarrhea, muscle cramps, fatigue, nausea, vomiting, insomnia, headache; common colds, anorexia, hallucinations, agitation, aggressive behavior, syncope, dizziness, abdominal disturbance, rash, pain, accident, abnormal dreams.
Drug Interactions
Ketoconazole, itraconazole, erythromycin, quinidine, fluoxetine. Rifampicin, phenytoin, carbamazepine, alcohol. Medications w/ anticholinergic activity, succinylcholine, other neuromuscular blockers, cholinergic agonists, β-blockers.
