Allopurinol
Indications/Uses
Clinical manifestations of urate/uric acid deposition. Management of 2, 8-dihydroxyadenine nephrolithiasis & recurrent mixed Ca oxalate nephrolithiasis. Hyperuricaemia secondary to malignancy.
Dosage/Direction for Use
Adult Mild 100-200 mg daily. Severe Max: 900 mg daily. Dose >300 mg should be administered in divided doses. Childn <15 yr 10-20 mg/kg daily or 100-400 mg daily.
Administration
Should be taken with food: Take immediately after meals.
Contraindications
Acute gout. Childn except those w/ hyperuricaemia secondary to malignancy. Asymptomatic hyperuricaemia.
Special Precautions
Hypersensitivity. Perform periodic hepatic & renal function tests & CBC. Ensure adequate hydration. Renal impairment. Pregnancy & lactation.
Adverse Reactions
Rash & hypersensitivity reactions (discontinue immediately), nausea, vomiting, diarrhoea, intermittent abdominal pain, fever/chills, arthralgia, cholestatic jaundice, eosinophilia, mild leucocytosis, leukopenia, ecchymosis, headache, myopathy, peripheral neuropathy, neuritis, paresthesia, somnolence.
Drug Interactions
Azathioprine, mercaptopurine, captopril, coumarin anticoagulants, cyclosporin, adenine arabinoside, drugs w/ uricosuric activity, chlorpropamide.
