Malarone

Atovaquone 250 mg, proguanil HCl 100 mg

Prophylaxis of P. falciparum malaria & treatment of acute, uncomplicated P. falciparum malaria.

Prophylaxis Adult 1 tab daily, 24 or 48 hr before entering a malaria-endemic area, continued during the period of stay, until 7 days after leaving the area. Treatment Adult 4 tab as single dose for 3 consecutive days. Childn >40 kg Same as adult. All doses to be taken as a single dose for 3 consecutive days.

Should be taken with food: Take at the same time each day w/ meals or a milky drink.

Hypersensitivity to atovaquone, proguanil or any excipients of Malarone. Prophylaxis of P. falciparum malaria in patients w/ severe renal impairment (CrCl <30 mL/min).

Not for the treatment of cerebral malaria or other severe manifestations of complicated malaria including hyperparasitaemia, pulmonary oedema or renal failure. Treat patient w/ different blood schizonticide if recrudescent infections due to P. falciparum or failure of chemoprophylaxis occurs. Parasites relapse w/ monotherapy for P. vivax. Additional treatment w/ a drug that is active against hypnozoites in intense exposure or malaria due to P. vivax & P. ovale. Repeat dose if patient vomits w/in 1 hr of dosing & continue to comply w/ personal protection measures (repellants, bednets). Consider alternative therapy in acute malaria w/ diarrhoea & vomiting or patients w/ severe renal impairment (CrCl <30 mL/min). Monitor parasitaemia w/ concurrent tetracycline. Discontinue if patients experience allergic reaction. Concomitant administration w/ rifampicin or rifabutin or metoclopramide is not recommended. Pregnancy & lactation. Childn <40 kg.

Headache, abdominal pain, nausea, vomiting, diarrhoea. Anaemia, neutropenia, rash, hyponatremia, anorexia, insomnia, dizziness, elevated liver enzyme levels, fever, cough, allergic reaction, abnormal dreams, depression, pruritus.

Warfarin & other coumarin-based anticoagulants, tetracycline, metoclopramide, rifampicin & rifabutin, efavirenz or boosted protease-inhibitors. Caution for patients receiving concomitant therapy w/ etoposide.
