Contents

Indications/Uses
FC tab/oral susp: Treatment of common bacterial infections where oral therapy is indicated: URTI (including ENT), skin & soft tissue infections, lower resp tract infections, GUT infections, osteomyelitis, septicaemia, peritonitis, post-op infections. 1,200 mg vial: Treatment of severe infections of ENT, acute exacerbation of chronic bronchitis, community acquired pneumonia, cystitis, pyelonephritis; skin & soft tissue, bone & joint, intra-abdominal, female genital infections. Prophylaxis in infections associated in major surgical procedures in adults.
Dosage/Direction for Use
FC tab Adult & childn >12 yr 1 g bd. Mild to moderate infections 375 mg tds. More severe infections, resp tract infections 625 mg tds. Oral susp Childn 7-12 yr 312.5 mg tds, 2-7 yr 156.25 mg tds, 9 mth to 2 yr 78.12 mg tds. 457 mg/5 mL oral susp Childn 2 mth-12 yr Mild to moderate infections 25/3.6 mg/kg/day. More severe infections 45/6.4 mg/kg/day. 1,200 mg vial Treatment of infections Adult & childn (wt >40 kg) 1,000 mg/200 mg slow IV inj every 8 hr. Surgical prophylaxis For procedures <1 hr in duration: 1,000 mg/200 mg to 2,000 mg/200 mg given at induction of anaesth. For procedures >1 hr in duration: 1,000 mg/200 mg to 2,000 mg/200 mg given at induction of anaesth, w/ up to 3 doses of 1,000 mg/200 mg in 24 hr. Childn (wt <40 kg): Childn ≥3 mth of age 25 mg/5 mg/kg every 8 hr. Childn <3 mth of age or weighing <4 kg 25 mg/5 mg/kg every 12 hr.
Administration
Should be taken with food: Take w/ meals for better absorption.
Contraindications
Hypersensitivity to amoxicillin, clavulanic acid, penicillins. Infectious mononucleosis, lymphatic leukemia. Caution in patients w/ allergic diathesis, bronchial asthma, urticaria hay-fever; babies born of mother hypersensitive to penicillin. History of liver problems or jaundice caused by penicillins or amoxicillin/clavulanic acid combinations.
Special Precautions
Hypersensitivity to other β-lactam antibiotics (eg, cephalosporins). Discontinue if allergic reactions occur. Patients w/ severe allergies or asthma; GI disturbances w/ vomiting &/or diarrhoea. Antibiotic-induced pseudomembranous colitis. Check hepatic & renal function & haematological status on long-term treatment. In the presence of bladder catheter, drink a lot of fluids to prevent possibility of crystalluria. Risk of superinfection w/ resistant bacteria or yeasts on long-term use. False +ve results in nonenzymatic methods for determining urinary sugars. Urobilinogen tests can be affected. +ve Coombs' test. Patients w/ severe renal or hepatic impairment. Pregnancy & lactation. Oral susp: Contains aspartame.
Adverse Reactions
Diarrhoea, nausea, vomiting, digestive disturbances; skin rash, pruritus, urticaria, fever, joint pain; leukopenia, thrombocytopenia, agranulocytosis, eosinophilia. Superinfections & colonisation w/ resistant organisms or yeasts; vag itching & discharge.
Drug Interactions
Increased MTX toxicity. Increased incidence of exanthema w/ allopurinol. Physically & chemically incompatible w/ aminoglycosides. Risk of bleeding w/ oral anticoagulants. May reduce efficacy of OCs. Reduced absorption of amoxicillin w/ antacids. Combination w/ rifampicin acts antagonistically. Do not concomitantly use w/ disulfiram. Avoid alcohol consumption during & for some days after treatment. Potential masking of effect of amoxicillin w/ chloramphenicol, macrolides, sulphonamides. Increased absorption of digoxin. May reduce plasma conc of sulfasalazine. Elevated serum conc of amoxicillin w/ probenecid.
