Indications and Dosage

Oral
Plaque psoriasis

Adult: As 4 mcg/g ointment or lotion: Apply sparingly onto affected area(s) once daily, preferably at bedtime. Max dose: 10 g ointment or 10 mL daily; 280 mcg per week (e.g. 30 mL lotion + 40 g ointment).

Contraindications

Hypercalcaemia, Ca metabolism disorder.

Special Precautions

Patient with generalised pustular or erythrodermic exfoliative psoriasis. Renal impairment. Pregnancy and lactation. Combined treatment with UV radiation.

Adverse Reactions

Significant: Hypercalcaemia.
Nervous system disorders: Paraesthesia.
Skin and subcutaneous tissue disorders: Skin irritation, itching, burning, erythema, contact dermatitis.

Patient Counseling Information

Avoid contact with eyes.

Monitoring Parameters

Monitor serum Ca level.

Drug Interactions

May increase the risk of hypercalcaemia with thiazide diuretics and vitamin D preparations. Increased therapeutic effect and shorten UV radiation period.

Action

Description: Tacalcitol, a synthetic vitamin D₃ derivative, is an antipsoriatic agent. It binds to vitamin D receptors to induce differentiation and inhibits hyper-proliferation of keratinocytes.
Synonym: tacalcitolum.
Pharmacokinetics:
Absorption: 0.5% absorbed through psoriatic skin.
Distribution: Completely bound to plasma proteins.
Excretion: Mainly via urine.
