Indications and Dosage

Oral
Gastrointestinal tract spasm

Adult: For the relief of smooth muscle spasms in conditions such as irritable bowel syndrome and diverticular disease of the colon: 60-120 mg 1-3 times daily. Consult the doctor if your symptoms worsen or do not improve after 2 weeks of treatment, or if you develop new symptoms.
Child: ≥12 years Same as adult dose.

Oral
Primary dysmenorrhoea

Adult: 60-120 mg 1-3 times daily. Consult the doctor if your symptoms worsen or do not improve after 2 weeks of treatment, or if you develop new symptoms.
Child: ≥12 years Same as adult dose.

Contraindications

Paralytic ileus, intestinal obstruction.

Special Precautions

Patient with nausea or vomiting, anorexia, weight loss, pallor, fatigue, fever, blood from the bowel, severe constipation, dysuria, and abnormal vaginal discharge or bleeding. Children. Pregnancy and lactation.

Adverse Reactions

Significant: Jaundice (reversible); abnormal LFTs.
Gastrointestinal disorders: Nausea.
Hepatobiliary disorders: Hepatitis (reversible).
Immune system disorders: Hypersensitivity reaction, anaphylaxis.
Nervous system disorders: Headache, dizziness.
Respiratory, thoracic and mediastinal disorders: Dyspnoea, wheezing.
Skin and subcutaneous tissue disorders: Rash, pruritus.

Patient Counseling Information

This drug may cause dizziness, if affected, do not drive or operate machinery.

Overdosage

Symptoms: Atropine-like effects and hypotension. Management: Supportive treatment for hypotension. Manage atropine-like effects as for atropine poisoning.

Action

Description: Alverine is an antispasmodic agent that directly acts on the intestinal and uterine smooth muscles, without affecting the heart, blood vessels and tracheal muscles.
Pharmacokinetics:
Absorption: Rapidly absorbed from the gastrointestinal tract. Time to peak plasma concentration: 1-1.5 hours (PHA).
Metabolism: Rapidly metabolised into para hydroxy alverine (PHA), primary active metabolite; undergoes further conversion to form 2 secondary metabolites.
Excretion: Via urine (as metabolites). Elimination half-life: 0.8 hours (alverine); 5.7 hours (PHA).
