Abacavir sulfate
Indications/Uses
Antiretroviral combination therapy for HIV in adult & childn.
Dosage/Direction for Use
Tab Adult, adolescent & childn from 3 mth at least 25 kg 300 mg bd or 600 mg once daily. Childn ≥20 kg to <25 kg ½ tab in the morning & 1 tab in the evening, 14 to <20 kg ½ tab bd. Oral soln Adult, adolescent & childn at least 25 kg 15 mL bd or 30 mL once daily. Childn <14 kg 8 mg/kg bd or 16 mg/kg once daily, up to max: 300 mg bd or 600 mg once daily. Mild hepatic impairment (Child-Pugh score 5-6) 200 mg (10 mL) bd.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Hypersensitivity. Discontinue use in patients w/ HLA-B* 5701 allele or if HSR is suspected. Lactic acidosis & severe hepatomegaly w/ steatosis. Immune reconstitution syndrome; MI. Opportunistic infections & other complications of HIV infection. Manage serum lipids & blood glucose. Known risk factors for liver disease. Avoid in patients w/ ESRD. Pregnancy & lactation. Mitochondrial dysfunction in neonates & infants exposed in utero or peri-partum to NRTIs. Childn <3 mth. Oral soln: Hereditary fructose intolerance.
Adverse Reactions
Anorexia; headache; nausea, vomiting, diarrhoea; fever, lethargy, fatigue.
Drug Interactions
Increased AUC by ethanol. Reduced Cmax & delayed Tmax w/ methadone.
