Ursodeoxycholic acid
Indications/Uses
Dissolution of radiolucent cholesterol gallstones in gallbladder <15 mm in diameter. Cholestatic liver disease (eg, compensated primary biliary cirrhosis, cholestasis of pregnancy).
Dosage/Direction for Use
Dissolution of cholesterol gallstones Approx 10 mg/kg daily equiv to: Up to 60 kg 2 cap, 61-80 kg 3 cap, 81-100 kg 4 cap, >100 kg 5 cap. Duration: 6-24 mth to dissolve. Cholestatic liver disease Approx 10-15 mg/kg equiv to: 34-50 kg 2 cap, 51-65 kg 3 cap, 66-85 kg 4 cap, 86-110 kg 5 cap, >110 kg 6 cap.
Administration
Should be taken with food: Swallow whole w/ some liqd.
Contraindications
Acute inflammation of gallbladder & bile duct, occlusion of biliary tract; calcified or X-ray dense gallstones (non-cholesterol gallstones), disturbed contractility of gallbladder, gallbladder filled >½ w/ gallstones, occlusion of ductus cysticus, frequent biliary colic.
Special Precautions
Monitor liver function parameters every 4 wk during 1st 3 mth, thereafter every 3 mth. Visualize gallbladder w/ overview & occlusion views in standing & supine positions 6-10 mth after beginning of treatment. Women of childbearing potential should use effective contraception during treatment. Not to be used during 1st trimester of pregnancy & lactation.
Adverse Reactions
Pasty stools or diarrhoea.
Drug Interactions
Reduced absorption by cholestyramine, colestipol or Al hydroxide-containing antacids &/or smectite (Al oxide). Increased absorption of cyclosporine. Reduced absorption of ciprofloxacin. Reduced AUC & Cmax of nitrendipine. Reduced therapeutic effect w/ dapsone.
