Indications and Dosage
Topical/Cutaneous
Aid to haemostasis when bleeding from capillaries and small venules
Adult: In cases where oozing blood and minor bleeding from capillaries and small venules is accessible and control of bleeding by standard surgical techniques (e.g. cautery, suture, ligature) is ineffective or impractical: As human-derived or recombinant solution: Apply directly or in conjunction with absorbable gelatine sponge onto the bleeding site. Dose depends on the size of the bleeding area. As bovine-derived solution: Apply 1,000-2,000 international units/mL for profuse bleeding or 100 international units/mL for bleeding from skin or mucosal surfaces. As bovine-derived powder: Apply directly onto the bleeding sites. Dosing recommendations and methods of application may vary among individual products (refer to specific product guidelines).

Contraindications
History of hypersensitivity to thrombin or other human blood products. Severe or brisk arterial bleeding treatment. Administration directly into the circulatory system. Bovine thrombin: Known or suspected antibodies to bovine thrombin and/or factor V.
Special Precautions
Children. Pregnancy and lactation.
Adverse Reactions
Significant: Hypersensitivity reactions (e.g. anaphylaxis), risk of infection transmission.
Blood and lymphatic system disorders: Anaemia, thrombocytopenia.
Investigations: Prolonged aPTT or prothrombin time, increased INR, increased neutrophil count, decreased lymphocyte count.
Cardiac disorders: Tachycardia.
Vascular disorders: Hypotension.
Potentially Fatal: Severe bleeding or thrombosis (bovine thrombin).
Monitoring Parameters
Monitor for signs of hypersensitivity, bleeding, thrombosis and abnormal coagulation laboratory values.
Action
Description:
Mechanism of Action: Thrombin is a protein substance produced from prothrombin. It catalyses the conversion of soluble fibrinogen into insoluble fibrin to promote haemostasis.
Storage
Store between 2-25°C. Do not freeze. Storage recommendations may vary among individual products (refer to specific product guidelines).
