Indications/Uses
In conjunction w/ other anti-TB drugs in the treatment of all forms of TB.
Dosage/Direction for Use
Adult & adolescent Prophylaxis: 300 mg once daily. Treatment: 300 mg once daily or initially 300 mg once daily for 2 mth then 15 mg/kg bd wkly for the remainder of the therapy. Max: 900 mg. Childn Prophylaxis: 10 mg/kg. Treatment: 10-20 mg/kg. Max: 300 mg once daily.
Contraindications
Hypersensitivity to INH, ethionamide, pyrazinamide, niacin (nicotinic acid) or other chemically related medications. Epileptics & patients w/ chronic alcoholism. Patients taking potentially hepatotoxic agents. Hepatic or renal impairment.
Special Precautions
Drug-induced liver disease. Patients suffering from convulsive disorder; DM. Pregnancy & lactation. Childn. Elderly >50 yr.
Adverse Reactions
Hepatitis, peripheral neuritis, blood dyscrasias, hypersensitivity, neurotoxicity, optic neuritis.
Drug Interactions
Increased incidence of INH-induced hepatotoxicity & increased metabolism w/ alcohol. Decreased plasma clearance & prolonged alfentanil duration of action. Induced microsomal metabolism by carbamazepine. Increased incidence of CNS effects w/ disulfiram. Increased potential for hepatotoxicity w/ other hepatotoxic medications & rifampicin. Decreased ketoconazole conc. Parenteral miconazole. Increased serum conc & inhibited metabolism of phenytoin.
