Ondansetron
Indications/Uses
Management of cytotoxic chemo- & RT-induced nausea & vomiting (CINV & RINV). Prevention & treatment of post-op nausea & vomiting (PONV).
Dosage/Direction for Use
CINV & RINV Adult 8 mg 1-2 hr prior to chemo- or RT, followed by 8 mg every 12 hr for max: 5 days. Highly emetogenic chemotherapy Single dose up to 24 mg w/ oral dexamethasone Na phosphate 12 mg 1-2 hr prior to chemotherapy. CINV Childn & adolescent 3-17 yr, >1.2 m2 5 mg/m2 or 8 mg IV + 8 mg tab after 12 hr on day 1 then 8 mg tab every 12 hr on days 2-6, ≥0.6 to ≤1.2 m2 5 mg/m2 IV + 4 mg tab after 12 hr on day 1 then 4 mg tab every 12 hr on days 2-6, >10 kg Up to 3 doses of 0.15 mg/kg IV every 4 hr on day 1 then 4 mg tab every 12 hr on days 2-6. Commence oral dosing 12 hr after IV dose, may be continued for up to 5 days. Total dose: Not to exceed 32 mg over 24 hr. Adult PONV 16 mg 1 hr prior to anaesth. Moderate or severe hepatic impairment Not to exceed 8 mg total daily dose.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Concomitant use w/ apomorphine.
Special Precautions
Hypersensitivity to other selective 5-HT3 receptor antagonists. Signs of subacute intestinal obstruction. Not recommended during pregnancy & lactation.
Adverse Reactions
Headache, sensation of warmth or flushing, hiccups, constipation.
Drug Interactions
Increased oral clearance & decreased blood conc w/ potent CYP3A4 inducers ie, phenytoin, carbamazepine, rifampicin. Reduced analgesic effect of tramadol.
