Piracetam
Indications/Uses
Cerebral functional impairment eg, loss of memory, lack of concentration or alertness & vertigo in the elderly. In combination w/ other antimyoclonic therapies for myoclonus of cortical origin, irrespective of aetiology.
Dosage/Direction for Use
Initially 7.2 g, increasing by 4.8 g every 3 or 4 days. Max: 24 g in 2 or 3 divided doses.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to piracetam or other pyrrolidone derivatives. Cerebral haemorrhage, patient suffering from Huntington's Chorea. Severe renal impairment (CrCl <20 mL/min); ESRD.
Special Precautions
Severe haemorrhage, risk of bleeding eg, GI ulcer; underlying haemostatis disorders, history of haemorrhagic CVA, undergoing major surgery & using anticoagulants or platelet antiaggregant drugs including low-dose aspirin. Avoid abrupt discontinuation. Patients on controlled Na diet. May affect ability to drive or operate machinery. Renal insufficiency. Not to be used during pregnancy & lactation. Long-term treatment in elderly.
Adverse Reactions
Nervousness; hyperkinesia; increased wt.
Drug Interactions
Confusion, irritability & sleep disorder w/ thyroid hormones. Decreased platelet aggregation, β-thromboglobulin release, fibrinogen & von Willebrand's factor levels & whole blood & plasma viscosity w/ acenocoumarol.
