Indications and Dosage
Nasal
Cranial diabetes insipidus
Adult: Per spray contains approximately 0.007 mg lypression (equivalent to 2 Posterior Pituitary Units): 1-2 sprays in each nostril 4 times daily.
Child: Per spray contains approximately 0.007 mg lypression (equivalent to 2 Posterior Pituitary Units): ≥6 wk: 1-2 sprays in each nostril 4 times daily.
Contraindications
Chronic nephritis with nitrogen retention.
Special Precautions
Hypertension or conditions which can be exacerbated by increased BP; nasal absorption may be impaired in rhinitis. Pregnancy.
Adverse Reactions
Nasal congestion, irritation, ulceration. Hypersensitivity reactions. Large doses: Marked pallor, pounding headache, vertigo, sweating, tremor, GI disturbances, eructation, cramp, desire to defaecate; hyponatraemia.
Drug Interactions
Antidiuretic effects increased with chlorpropamide, clofibrate, carbamazepine, fludrocortisone, urea, TCAs. Effects reduced with lithium, heparin, demeclocycline, noradrenaline, alcohol. Ganglion-blocking agents may enhance sensitivity to pressor effects of lypressin.

Description:
Mechanism of Action: Lypressin is a synthetic polypeptide with vasoconstrictor and antidiuretic effects.
Onset: Nasal route: Within 1 hr; time to peak: 30-120 minutes.
Duration: Nasal route: 3-4 hr.
Pharmacokinetics:
Absorption: Rapid from the nasal mucosa.
Metabolism: Renal and hepatic pathways.
Excretion: Half-life: About 15 minutes.
Storage
Store between 15-30°C.
