Granisetron HCl
Indications/Uses
Prevention & treatment of acute & delayed chemotherapy- & RT-induced nausea & vomiting (CINV & RINV); post-op nausea & vomiting (PONV).
Dosage/Direction for Use
Adult CINV & prevention of RINV 1-3 mg slow IV inj over 30 sec or IV infusion over 5 min prior chemo- &/or RT. CINV Max dose: 9 mg over 24 hr. PONV prevention 1 mg slow IV inj over 30 sec prior to anesth induction. PONV treatment 1 mg slow IV inj over 30 sec. Max dose: 3 mg daily. Ped CINV 10-40 mcg/kg IV infusion over 5 min prior to chemotherapy, may administer additional dose w/in 24 hr at least 10 min after initial dose.
Contraindications
Special Precautions
Signs of subacute intestinal obstruction, cardiac co-morbidities. Patients on cardiotoxic chemotherapy &/or w/ concomitant electrolyte abnormalities. Concomitant use w/ other serotonergic drugs. May affect ability to drive. Pregnancy & lactation.
Adverse Reactions
Constipation; headache, agitation, anxiety, CNS stimulation, dizziness, insomnia, somnolence; fever; HTN; transient increase in AST & ALT; skin rashes; taste disorder.
Drug Interactions
Induced metabolism w/ phenobarb. Increased apomorphine levels/effect. Serotonin syndrome w/ other serotonergic drugs. Drugs prolonging QT interval &/or are arrythmogenic.
