Ademetionine
Indications/Uses
Intrahepatic cholestasis in pregnancy & in precirrhotic & cirrhotic states. Relief of fatigue caused by chronic liver disease.
Dosage/Direction for Use
Adult Intrahepatic cholestasis Recommended dose: 10-25 mg/kg daily. Usual starting dose: 800 mg daily. Total daily dose: 1,600 mg. Maintenance: 800-1,600 mg daily.
Administration
Should be taken on an empty stomach: Swallow whole, do not chew/crush.
Contraindications
Special Precautions
Monitor ammonia levels in patients w/ hyperammonemia in precirrhotic & cirrhotic states. Routine blood test to check plasma levels of ademetionine in patients at risk of vit B12 & folate deficiencies. May affect ability to drive or operate machinery. Renal impairment. Pregnancy & lactation. Childn. Elderly.
Adverse Reactions
UTI; confusion, insomnia; dizziness, headache, paresthesia; CV disorder; hot flush, superficial phlebitis; abdominal distension & pain, diarrhea, dry mouth, dyspepsia, esophagitis, flatulence, GI pain & hemorrhage, nausea, vomiting; biliary colic, hepatic cirrhosis; hyperhydrosis, pruritus, skin reactions; arthralgia, muscle spasms; asthenia, chills, inj site reactions, flu-like illness, malaise, peripheral edema, pyrexia.  
Drug Interactions
Serotonin syndrome w/ clomipramine. SSRIs, TCAs, tryptophan-containing over-the-counter drugs & herbal supplements.
