Per tab Co-trimoxazole: Sulfamethoxazole 400 mg, trimethoprim 80 mg. Per 5 mL oral susp Co-trimoxazole: Sulfamethoxazole 200 mg, trimethoprim 40 mg
Indications/Uses
Upper & lower urinary tract, resp & genital tract, GIT, skin & wound infections. Bacterial infections caused by sensitive organisms, fungal skin infections. South American blastomycosis.
Dosage/Direction for Use
Tab Acute infection Adult & childn >12 yr 2 tab every 12 hr. Childn 6-12 yr 1 tab every 12 hr. Oral susp Childn 6-12 yr 10 mL every 12 hr, 6 mth-5 yr 5 mL every 12 hr, 6 wk-5 mth 2.5 mL every 12 hr. Acute infections Duration: At least 5 days. Acute uncomplicated lower UTI Short-term therapy: 1-3 days. Usual duration: 10-14 days. Shigellosis Duration: 5 days. CrCl >30 mL/min Usual dosage, 15-30 mL/min ½ usual dosage.
Administration
Should be taken with food.
Contraindications
Hypersensitivity to sulfonamide or trimethoprim. Marked liver parenchymal damage. Not to be given to patients w/ serious haematological disorder. G6PD deficiency. Severe renal insufficiency. Pregnancy. Premature & newborn babies during 1st wk of life.
Special Precautions
Discontinue use if skin rash appears. Folic acid deficiency; blood dyscrasias; G6PD deficiency; severe allergy or bronchial asthma; group A β-haemolysis streptococcal tonsillopharyngitis. Superinfection. Ensure adequate fluid intake. Monitor blood count regularly on long-term therapy. Renal & hepatic impairment. Not to be given during pregnancy. Lactation.
Adverse Reactions
GI disturbances eg, nausea, vomiting, anorexia; skin rashes; glostitis, pancreatitis, stomatitis, diarrhoea.
Drug Interactions
Increased thrombocytopenia w/ diuretics. Megaloblastic anaemia w/ pyrimethamine. Potentiated activity of oral hypoglycaemic or anticoagulants. Displacement of warfarin. Inhibited hepatic metabolism of phenytoin. Shortened plasma t½ w/ rifampicin.
