Contents
Amoxicillin 500 mg, clavulanic acid 125 mg
Indications/Uses
Upper & lower resp tract (including ENT), skin & soft tissue, GUT, bone & joint infections; intra-abdominal sepsis.
Dosage/Direction for Use
Adult & childn >12 yr 625 mg bd. Max duration: 14 days. Dental infections 625 mg tab bd for 5 days. CrCl 10-30 mL/min 625 mg bd. CrCl <10 mL/min Not >375 mg bd.
Administration
May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort. May be divided in half & swallowed whole w/o chewing.
Contraindications
Hypersensitivity to penicillin & cephalosporins. History of jaundice/hepatic dysfunction associated w/ co-amoxiclav.
Special Precautions
Hypersensitivity reactions to penicillin, cephalosporins or other β-lactam agents; anaphylactoid reactions. Avoid use if infectious mononucleosis suspected. Overgrowth of nonsusceptible organisms. Atopic individuals. Concomitant use w/ allopurinol, anticoagulant therapy. Maintain adequate fluid intake & urinary output. Regularly check patency in patients w/ bladder catheters. Prolonged use. Severe hepatic dysfunction, cholestatic jaundice. Moderate or severe renal impairment. Avoid use during pregnancy & lactation. Not suitable in childn <12 yr.
Adverse Reactions
GI disturbances; hypersensitivity reactions; moderate increased AST (SGOT) &/or ALT (SGTP) levels, hepatic dysfunction including increased serum transaminase, serum bilirubin &/or alkaline phosphatase; anemia, thrombocytopenia, thrombocytopenic purpura, eosinophilia, leucopenia, agranulocytosis.
Drug Interactions
Increased INR ratio w/ oral anticoagulants. Increased & prolonged blood levels of amoxicillin w/ probenecid. Lowered oestrogen reabsorption. Reduced efficacy of combined OCs.
