Contents
Per 625 mg tab Amoxicillin 500 mg, clavulanic acid 125 mg. Per 1,000 mg tab Amoxicillin trihydrate 875 mg, clavulanic acid 125 mg. Per 31.25 mg/mL powd for oral susp Amoxicillin trihydrate 25 mg, clavulanic acid 6.25 mg. Per 1.2 g powd for inj Amoxicillin Na 1,000 mg, clavulanic acid 200 mg
Indications/Uses
Short-term treatment of upper & lower resp tract, GUT, skin & soft tissue, bone & joint, & dental infections. Septic abortion, puerperal & intra-abdominal sepsis. Prophylaxis of wound infection associated w/ surgical procedures.
Dosage/Direction for Use
Tab Adult, childn >12 yr & mild renal impairment (CrCl >30 mL/min) Mild to moderate infections 625 mg bd or 375 mg tds. Severe infections 1,000 mg bd. Dental infections 625 mg bd for 5 days. CrCl <10 mL/min Not >625 mg every 24 hr. Powd for oral susp Childn 7-12 yr 10 mL tds, 2-7 yr 5 mL tds, 9 mth to 2 yr 25 mg/kg daily in 3 divided doses. Powd for inj Adult & mild renal impairment (CrCl >30 mL/min) 1.2 g every 8 hr by IV inj or infusion, may be increased to every 6 hr in more serious infections. Childn 3 mth-12 yr 30 mg/kg every 8 hr, may be increased to every 6 hr in more serious infections, 0-3 mth 30 mg/kg every 12 hr, may be increased to every 8 hr thereafter. Surgical prophylaxis 1.2 g at induction, may be given further 2-3 doses every 8 hr in 24-hr period for high-risk procedures. Moderate renal impairment (CrCl 10-30 mL/min) 1.2 g IV stat, followed by 600 mg IV every 12 hr, severe renal impairment (CrCl <10 mL/min) 1.2 g IV stat, followed by 600 mg IV every 24 hr; additional 600 mg IV may need to be given during & at the end of dialysis.
Administration
May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort. FC tab: May be broken in half & swallowed whole w/o chewing.
Contraindications
Hypersensitivity to β-lactam antibiotics. Penicillin-associated cholestatic jaundice or hepatic dysfunction.
Special Precautions
Hypersensitivity to penicillins & cephalosporins. Reduced urine output, erythematous rashes associated w/ glandular fever. Ensure adequate fluid intake especially during high doses. Avoid prolonged use. Cholestatic jaundice. Renal or hepatic impairment. Avoid use in pregnancy.
Adverse Reactions
Diarrhoea. Mucocutaneous candidiasis; nausea, vomiting; urticarial & erythematous rashes, angioedema, oedema, anaphylaxis, serum-sickness like syndrome, hypersensitivity vasculitis; thrombophlebitis at inj site.
Drug Interactions
Prolonged bleeding & prothrombin time w/ anticoagulants. Reduced efficacy of OCs. Decreased amoxicillin renal tubular excretion w/ probenecid. Increased likelihood of allergic skin reaction w/ allopurinol.
